On the Temple Mount / Noble Sanctuary in Jerusalem, this building (along with the nearby al-Aqsa Mosque) is the third holiest in Islam (after the Ka'ba in Mecca and the Prophet's -- saw -- Mosque in Medina).
Muslims believe that the rock inside the building is the location from where Muhammad ascended to heaven one night, while meeting a number of prophets along the way. There is a story that when Muhammad was rising up, the ground wanted to rise with him, but the Angel Gabriel told the ground its place was on Earth and so a small cave was created -- that cave is still existant today inside the Dome.
To the Jews, the foundation stone of the Temple Mount, above the cave, is the holiest site in Judaism along with the nearby Western Wall. They face towards it everyday while in prayer.
